efficiency是多少？很多paper里就给了一个Immunostaining的图，也没给个数字。根
据大家的经验，这个效率大概是多少呢？（如果能讨论下特异性就更好了）
另外，liver大概有70%的细胞是hepatocytes，剩下的就是其他细胞了，transfection
是针对所有的细胞吗？有没有特异性的针对hepatocytes的。

epithelial cells, smooth muscle cell and more 2),the cells must form a 3D structure
that accompany tons of sinus, boold vessels and bile ducts. That means all
cells must coordinate precisely knowing where, when to grow and to stop. In
the embryonic stage, embryos have many factors to guid the organogenesis,
but in adult stage, those factors are not conveniently available any more.

患者44岁，男，6年前因为肝癌做了肝移植 （符合米兰标准），之后无灾无病好几年。
去年2016年初发现轻微贫血，血色素12，因为当时在服小剂量阿司匹林，于是停服，看
看有没好转。托托拉拉到了年底，还是贫血，血色素10.5。医生说做个内镜。于是在胃
内发现一个肿瘤，内镜医生直觉是GIST。随后CT，但CT却没有异常。4月初第二次胃镜
，GIST依然还在，取样活检表明可能肝癌转移或者是非常罕见的HAC。第二次CT 报告大
小46x35mm，说比较第一次32mm变大了。可能第一次看片医生那天喝多了？肿瘤位置在
Fundus，非常接近GOJ。
AFP好久没有查了，因为肝上的情况一直很好，肝移植前AFP 100多，移植后AFP一直小
于2。上一次至少是2，3年前。也是疏忽了。现在AFP 254. 显然胃上这个要么是转移，
要么是HAC。
下周二医生还要安排一次PETCT，周三见外科医生，估计排除转移外就安排切除了。 求
这里的专家教授指点，这个情况如何是好？下面我贴上最近几个报告。

的一项调查显示，88％的科学家认为转基因食品可安全食用，持这一看法的公众只有37
％。报告认为差距如此之大的一个可能原因是，三分之二的公众认为科学家对转基因食
品的健康后果没有清晰认识。这就是真正的原因吗？
根据报告全文，皮尤中心给19，984名科学家发出了调查问卷，3748人（18.8%）完成了
问卷调查。其中生物医学学科背景的科学家占50%。报告没有说明50%的生物医学背景是
否能够代表科学家总体的学科构成。更大问题是在这50%的科学家人群中，有多少人的
工作是与转基因作物的研究有关，以什么样的方式相关。报告没有提供任何信息。
由于这些关键信息缺失，88%的科学家相信转基因安全的说法毫无意义。在一个没有代
表性并且无法排除偏向性的样本中调查，科学家相信转基因安全从1%到99%与88%没有任
何不同。好消息是超过半数的美国民众认为转基因不安全。

details.
Here is what I used for hepatocyte.
Preparation of the fatty acid stocks
The free fatty acids are prepared by dissolving each individual free fatty
acid in 100% ethanol. Most are readily dissolved in ethanol, though some
require heating to achieve fully solubility.
A final concentration of 100 mM for each of the fatty acids as follows:
31 ul 1 M Palmitic Acid Solid, 1 g / 3.3 ml EtOH

发帖数: 351 | 47 这几种脂肪酸中，Palmitic Acid 和 Stearic Acid常温下是固体，计算摩尔浓度比较
容易。其他几种常温下是液体，但是大体上他们的密度和水的密度很相近，1ml近似1g
。这些脂肪酸都极易溶于乙醇。溶解Palmitic Acid需要略微加热（50C)。
The free fatty acids are toxic unless they are presented bound to purified,
fatty acid free, endotoxin-free serum albumin, which is used at a typical
concentration of 0.1–0.2%.
Take 31ul 1M palmitic acid, 2.8ul 1M Palmitoleic acid etc and bring the
final volume to 1000ul with 100% EtOH, you will get 0.1M FFA. Add 76ul of

发帖数: 1704 | 48 这种类型研究的互相矛盾已经屡见不鲜了，基于clinical association的分析很容易引
入bias，而所谓的statistical significance有时候也不能说明问题。究竟有没有关联
HBV在其中又实际起了什么作用，很难说。
目前HBV在HCC中的作用都还停留在“假说”阶段，更别提什么pancreatic cancer了。
HBV的体外复制是严格肝细胞特异性的，而体外感染更是只能在primary hepatocytes上
实现（HepRG cell是一个例外），体内也未见有HBV感染pancreatic cell的报道。
假如HBV真的与pancreatic cancer相关的话，我个人倾向于认为，同样是基于某些间接
因素，编码产物直接作用于pancreatic cell，可能性很小

那些个receptor，没有一个是真正HBV specific甚至hepatocyte specific的，所以我
猜测这些不会是很好的target。PreS1 derived peptide作为entry inhibitor目前据说
效果已经非常好了，另有其他一些entry inhibitory peptide/molecule在pre-
clinical阶段的效果也不错，所以我估计眼下针对“疑似”receptor的rational
design不会有太大的吸引力。
chronic HBV治疗，要说最终解决还是得靠免疫学的方法，传统的NRTI也好，entry
inhibitor也罢，治标不治本，解决不了问题。强效Interferon以及前几天热炒的
Adoptive T cell therapy，我觉得应该是目前看来最有前景的方向吧，这玩意在国内
时市场太大了。

带者呢。。。国内还是要靠疫苗外加小分子药物。而小分子药物最直接的就是block病毒
entry.
当然了，你可以说乙肝携带者其实大多数的肝细胞都已经被侵染了 阻止entry用处不大
。但是现在乙肝在肝细胞之间怎么transfer的似乎了解的不是很清楚，也许阻止entry会
有意想不到的效果？（比如说，virus transfer对维持病毒的存在是关键的）
