经过一个中间宿体性的其它动物才能传染给人。所以，病毒发生后，就故意误导全国人
民去海鲜市场找证据，找其它野生动物的麻烦。 而且还是病毒所去找的，找完了还装
模做样化验呀，分离呀什么的。最后把责任全部推给了海鲜市场的动物。可是那种动物

C8H12O4和C4H8N6O2。
如果看fragmentation，所有的fragment按照N规则，似乎都是不含N的，要所有的
fragment都不含N，那么分子本身含N的可能性比较小。也就是说第二个的可能性比较小
。如果你的isotopic peak的ratio是准的话，和第二个分子式match，这样又排除了第
一个。而且第二个分子式算出来的mass和你测得的只差1.7个ppm，对于FTMS来说比较
make sense。而第一个分子式的计算值和你测得要差17.3个ppm，对于FTMS来说有点太
大了。
感觉分子式C4H8N6O2，但是回到开始说的N规则，如果有6个N的话，你所有的
fragmentation的碎片里面either要含有所有的6个N，或者每次掉偶数个N。另外看你的
fragmentation，碎片几乎都分布在比较靠近molecular ion的一端，低mass几乎看不到
太多的fragmentation，感觉最大的可能是你的6个N都不太容易掉，比如都长在几个共
轭的环上，环上有些支链。但你又说是个直链的东西，那可能是一些直链 |

size of fragments can joint together easier than different size fragments.

fragmentation有一定的pattern，所以产生的abcxyz fragment ion可以和从database
sequence中计算预测的fragment ion进行比较：能match的fragment越多，对这个
peptide的confidence就越高。
目前主流仪器对单个peptide的fragments能cover八九成，但在较复杂的体系中对不同
peptide的coverage会下降，这个主要还是仪器扫描速度，频率和通量等的限制，并不

There is two libraries i am going to prepare:
1), Regular fragmentation, 150 bp / fragment, then link to adaptors.
2), Mate-paired: link two ends of a fragment (1-2 kb) together. (i had
thought that is why also called 'paired-end') .
According to your description, pair-ends is the regular fragments, but why
was call 'pair-ends', is that because it was sequenced from two ends?

同一抗体IP这两个fragments， 然后再用mass spectrometry ID证实是目标蛋白的
fragments。
但目前IP后fragments的信号被igG heavy and light chains所屏蔽（both for IB and
MS).大家有什么办法可以去除IgG的信号？谢谢 |

是pUC19(SacI)-(SacI)fragment I(XmaI)-(XmaI)fragment II(BamHI)-(BamHI)
fragment III(XbaI)-(XbaI)fragmentIV(HindIII)-pUC19(HindIII)
每个fragment大概0.8-1.1kb
然后我做的是pUC19 50 ng,
pUC19:f1:f2:f3:f4=1:5:5:5
总体积15-20 ul
ligate 20 hrs
然后3 ul加到一管TOP10电感化态细胞里
涂100-200 ul/平板
然后挑克隆
用colony digestion筛选
一起做了五个ligation
一天筛了近90个克隆
每个ligation都筛出正确的了
然后这些positive的提质粒出来
用不同的酶切再确认一下
然后测个序（或者不测）
直接PCR的切就行
不过我个人比较喜欢的是每个片段先做到TOPO里面（pBluntII)
先测序
挑序列正确的再酶切、胶纯化，往下做
这样每一步都是正确的序列
最后终产物酶切什么都对的话不测序也没关系

CID在ION TRAP里还是最快的
FUSION叫TRIBRID是你可以同时做3件事情.我觉得如何利用这个优势更重要.比如在
ORBITRAP做MS1或者HCD的时候别让ION TRAP闲着.
CID FRAGMENT的时候是M/Z FOCUS的,意味着如果PEPTIDE上的修饰容易掉的话,就不会有
很好的BACKBONE FRAGMENTS (因为修饰掉了M/Z就变了), ETD可以解决这个. 但现在有
HCD了,是QUADRUPOLE FRAGMENTATION (修饰掉了还可以继续FRAGMENTATION),对修饰的
PEPTIDE效果也不错.
尤其FUSION的速度太快了,15HZ ORBITRAP, 20HZ ION TRAP. 那这个当QE使唤有些大材
小用了. |

发帖数: 728 | 34 如果你的peptide比较长，以至于产生很多+3--+5甚至更高的fragment ions，那么无论
对于ETD还是CID，在OT里做high resolution的MS2都更有利。而ETD是有利于high
charge state，long peptide的fragmentation。所以，如果你的样品确实有很多long
peptide的话（你去raw data file，average一段MS1的data就有底了），用OT scan
MS2会让搜索效果更好。你搜索时记得把fragment ions的error window舍得很窄，这样
才起到high resolution的效果。比如10 or 20ppm，或者0.01 m/z，取决于你具体的
accuracy。你可以试试MS2用15000 resolution with 20 ppm error window。
关于省时间的问题。诚然IT确实比OT省时间，如果你用老一代的LTQ Orbitrap，这个确
实是个大concern。但技术在进步，Orbitrap Fusion已经在设计上提供了比过去快得多

（1）卫生部2007年颁布实施《新资源食品管理办法》有意取消了2002年颁布实施
的《转基因食品卫生管理办法》“第五条 转基因食品的食用安全性和营养质量不得低
于对应的原有食品”这个条款，是退步，而不是进步！这种删除的实质是，进一步顺从
邪恶孟山都为首跨国转基因产业的私利与压力，置中国人民持续安全、健康、生存与繁
衍于不顾！
（2）卫生部2007年颁布实施《新资源食品管理办法》有意取消了2002年颁布实施
的《转基因食品卫生管理办法》“第五条”，但是卫生部《新资源食品管理办法》“第
三条”规定：
第三条 新资源食品应当符合《食品卫生法》及有关法规、规章、标准的规定，对
人体不得产生任何急性、亚急性、慢性或其他潜在性健康危害。
依据该第三条规定，在对于抗草甘膦转基因大豆加工的转基因大豆油，检测出超过
俄罗斯对大豆油草甘膦残留量允许标准与中国卫生部对棉籽油草甘膦残留最高限量2.9-
6倍的草甘膦残留量，已经证实对人类健康，特别对青少年、儿童、婴儿与孕妇及其胎

初速方面，看你准备用啥弹药. 小口径高初速弹讲究fragmentation, m855的reliable/
minimum fragmentation初速是2700-2500fps
16寸枪管， m855弹的fragmentation距离是大概100/150米，
14.5寸枪管则是50/100米。
14.5如果想可靠的让m855弹形成破片， 距离必须在50米内...这有点悲剧， 如果用
Hornady 75gr TAP, 破片距离是2250fps, 也就是175m
16寸枪管打75gr TAP则是200m
结论， SHTF只能大量囤积普通弹药， 所以要用16寸。 |

SE鸟山求：TBC的意味在于游戏结尾后还有很多后续内容，比如碎片结局，后续DLC等，所以没有用系列以往一贯用的“END”。
问：前传小说《THE FRAGMENTS BEFORE》最后也是TBC（TBC IN THE FRAGMENTS AFTER）呢。
SE鸟山求答：在后续的外传小说《THE FRAGMENTS AFTER》里会描写游戏中没有
交代的同时期发生的事情，比如比如解释雪叔怎样又变成路西等等。
问：游戏里打了几瓶酱油的黑人父子，详情完全不明了呢。
SE鸟山求：确实阿。比如黑人父子和鸟娘等事件（注：这三个是有关系的），目前在考虑是出DLC呢还是出小说等来补完。

问：雪叔为什么能穿越？他手上的路西印记是怎么回事？
SE渡边大佑：雪叔终于相信了赛拉的想法，于是独自寻找L姐。后来他找到并主动接近了一个法尔希，希望能从它身上得到穿越的能力，后来他就变成了路西并找到了一个门，于是就开始了他漫长的旅程，这段会在小说里描写。而接下来的故事在游戏里以文字的形式详细地表述了（英雄的轨迹）。

发帖数: 1027 | 45 隐藏结局要收集全160个fragment，我第一次通关还不到50个fragment。
大部分的fragment都是透明的，不看攻略能找得到吗？即便收集全了，隐藏结局的动画也
只有1分钟，而且和普通结局没差别啊，goddness挂了，对吧。
再吐槽一下小游戏，转钟表这个太无耻了吧，backtrack算法解很合适，人脑跟不上吧。
智力问题也很无耻，庞大的题库，连续10道答对才有奖品。
游乐场的公鸡赛跑很有趣？
最后再吐槽下PS3的FF13_2的save editor，没有Xbox版的好，只能批量修改最大化属性值

Hammadi)镇被发现，采用一种今日称之为科普特钉装法的方式来钉装。它与正典的四福
音不同之处在於：它除了声称是由耶稣十二门徒之一的多马(天主教作「多默」)所写以
外，全卷只是记录了耶稣的言论，并没有提及耶稣的传道过程也没有记载耶稣受难复活
的经历。
当本经文的完整科普特语版本译出後，圣经考古学家立即意识到其中的文句对应於1896
年到1904年间在奥克西林库斯（Oxyrhynchus）出土的希腊文残页(即写在残破蒲草纸板
的希腊文版多马福音)。根据考古学家的推断，目前所收集到的希腊文手抄版本来自三
份抄本（P. Oxy. I 1; IV 654; IV 655），都是在公元200年初所抄写，而这份科普特
语手抄版本则成卷於公元340年左右。虽然科普特语版本的内容与希腊文的残本并不完
全相同，但相信它是从更早期的希腊文古文翻译而来的。
